He studied the History and Literature of Russia and Britain at Harvard University afterward in 2000, where he became the president of Harvard Lampoon, the University's humor publication. It seems he showed early signs of entering the comedy and writing world as he and his friends reenacted SNL sketches and read David Letterman Top 10 Lists while in Regis High, under teacher's permission.

NBC's SNL has been the biggest and the first platform for Jost to enter the spotlight. For the money part, he won $5,250 on a college edition of the game-show, Weakest Link. Immediately after graduation from Harvard in 2004, he was hired as a writer for SNL.

Jost's job in SNL surely has toggled over the years on the show. After working as one of the writers of 102 episodes from 2005 to 2012, he was promoted as one of the head writers in 2012. He was already promoted to writing supervisor in 2009 by then. He took a hiatus from SNL writing in 2015 but returned as a co-head writer of the show in 2017. He's been on the writing staff of SNL for 290 episodes, as of December 20, 2019.

As for his on-screen appearances, he started working as the Weekend Update Anchor since 2014, while declaring the hiatus, for 123 episodes till now. He was also featured in earlier episodes of the show, just as an uncredited character.

For the anchor role, he was approached by executive producer Lorne Michael to replace Seth Meyers, who was moving on with his own late-night talk show, Late Night with Seth Meyers, in 2014.
