ECL is considered as the experts’ detection method of choice due to its high sensitivity, excellent signal-to-noise ratio, and wide dynamic range. Additionally, ECL is also useful in quantifying a wide variety of biological materials such as cell RNA, DNA and other analytes.
What does the ECL do in Western blotting?
The ECL Western Blotting Substrate detects picogram amounts of antigen, and with the use of photographic or other imaging methods, visualizes the presence of HRP. Blots can be repeatedly exposed to X-ray film to obtain optimal results or stripped of the immunodetection reagents and reprobed.
How do you apply ECL to membrane?
Drain the membrane of wash buffer. Position the membrane on a piece of Saran Wrap and quickly combine the following in chronological order while stirring: 10 ml ECL buffer (see Materials and Reagents), 22 µl p-coumaric acid, 50 µl luminol and 3 µl H2O2. Pour onto membrane and allow to stand undisturbed for 90 sec.
