Setting of agar plates: Agar plates when poured should be allowed to set undisturbed at room temperature. It is best practice that when completely set and cooled to room temperature that they should be stored stacked upside down in sealed plastic bags in the fridge at 4 °C.
How do you store inoculated agar plates?
Store agar plates in a refrigerator. Most bacteria cannot grow well in cold temperatures. Store plates in a cold room if a refrigerator is not available. If you are storing plates in a cold room, check the plates for condensation a few hours after pouring.
Where should inoculated agar be stored?
The plate should either be used or put into long term storage just before the mycelium reaches the edge of the plate. Otherwise, the mycelium could easily grow though the parafilm, inviting contamination. Most cultures can be stored long term in the refrigerator.
